MMR (Measles, Mumps, and Rubella) Vaccines

Measles, Mumps, and Rubella are three highly contagious viral diseases that can lead to serious complications such as pneumonia, encephalitis, infertility, or congenital rubella syndrome in pregnant women.

The MMR vaccine provides safe, long-term protection against all three diseases in a single injection and remains one of the most effective and essential immunizations recommended worldwide.

A. Priorix® (GlaxoSmithKline – GSK)

Formulation:

Live attenuated measles (Schwarz strain), mumps (RIT 4385 strain derived from Jeryl Lynn), and rubella (Wistar RA 27/3 strain) viruses, each propagated in chick embryo or human diploid (MRC-5) cell cultures. Lyophilized for reconstitution.

Packaging:

Single-dose vial (lyophilized powder) with pre-filled syringe of sterile water diluent, yielding 0.5 mL after reconstitution.

Dosage and Administration:

  • Children (≥ 12 months): One 0.5 mL subcutaneous dose; a second dose recommended at least 4 weeks later.
  • Adults: Two doses, 0.5 mL each, at least 4 weeks apart for full protection.

Indications:

Active immunization against measles, mumps, and rubella in individuals 12 months of age and older.

Contraindications:

  • Hypersensitivity to neomycin or any vaccine component.
  • Immunodeficiency or immunosuppressive therapy.
  • Pregnancy (avoid conception for one month post-vaccination).
  • Defer during febrile or acute illness.

Storage / Cold-Chain Requirement:

  • Store between +2°C and +8°C.
  • Do not freeze.
  • Protect from light; use immediately after reconstitution.

B. MMR-II® (Merck Sharp & Dohme – MSD)

Formulation:

Live attenuated measles (Enders Edmonston strain), mumps (Jeryl Lynn [B] strain), and rubella (Wistar RA 27/3 strain) viruses, lyophilized and supplied with diluent.

Packaging:

Single-dose vial (lyophilized vaccine) with pre-filled syringe of diluent, 0.5 mL after reconstitution.

Dosage and Administration:

  • Children (≥ 12 months): One 0.5 mL SC dose; second dose after 4–6 weeks.
  • Adults: Two doses, 0.5 mL each, separated by at least 4 weeks.

Indications:

Active immunization against measles, mumps, and rubella in persons 12 months and older.

Contraindications:

  • Hypersensitivity to neomycin or any component, including gelatin.
  • Immunocompromised individuals.
  • Pregnancy (avoid for one month post-vaccination).
  • Delay vaccination if febrile or moderately ill.

Storage / Cold-Chain Requirement:

  • Maintain +2°C to +8°C.
  • Do not freeze reconstituted vaccine.
  • Protect from light and administer immediately after mixing.
