Chickenpox (Varicella) Vaccines

Chickenpox, or Varicella, is a highly contagious viral infection caused by the Varicella zoster virus (VZV). While often mild in children, it can lead to severe complications in adults, pregnant women, and immunocompromised individuals—including pneumonia, skin infections, and hospital confinement.

Vaccination provides safe, long-lasting protection against varicella and helps reduce transmission in workplaces, schools, and households.

A. Varilrix® (GlaxoSmithKline – GSK)

Formulation:

Live attenuated Varicella zoster virus (Oka strain), lyophilized for reconstitution.

Packaging:

Single-dose vial (lyophilized powder) with pre-filled syringe of diluent, 0.5 mL after reconstitution.

Dosage and Administration:

  • Children (≥ 12 months): One 0.5 mL dose subcutaneously; a second dose recommended after at least 6 weeks for full protection.
  • Adolescents and adults (≥ 13 years): Two doses, 0.5 mL each, at least 4–8 weeks apart.

Indications:

Active immunization for the prevention of varicella infection in individuals aged 12 months and older.

Contraindications:

  • Hypersensitivity to neomycin or any vaccine component.
  • Immunodeficiency or immunosuppression (e.g., leukemia, lymphoma, HIV with severe immunosuppression).
  • Pregnancy — avoid vaccination during pregnancy and up to one month after.
  • Moderate or severe acute illness (postpone until recovery).

Storage / Cold Chain Requirement:

  • Store between +2°C and +8°C.
  • Do not freeze the reconstituted vaccine.
  • Protect from light.
  • Use immediately after reconstitution.

B. Varivax® (Merck Sharp & Dohme – MSD)

Formulation:

Live attenuated Varicella zoster virus (Oka/Merck strain), lyophilized for reconstitution with sterile diluent.

Packaging:

Single-dose vial (lyophilized vaccine) with pre-filled syringe of diluent, 0.5 mL dose after reconstitution.

Dosage and Administration:

  • Children (12 months to 12 years): One dose, 0.5 mL SC; second dose recommended after 3 months.
  • Adolescents and adults (≥ 13 years): Two doses, 0.5 mL SC each, 4–8 weeks apart.

Indications:

Active immunization for the prevention of varicella infection in individuals 12 months of age and older.

Contraindications:

  • Hypersensitivity to any component (e.g., neomycin, gelatin).
  • Immunocompromised individuals or those receiving immunosuppressive therapy.
  • Pregnancy (avoid conception for one month post-vaccination).
  • Defer in moderate or severe acute illness.

Storage / Cold Chain Requirement:

  • Store between +2°C and +8°C.
  • Do not freeze the reconstituted vaccine.
  • Protect from light.
  • Administer immediately after reconstitution.
